Hi everyone,
is anyone else considering taking part in any clinical trials? As I’m heading down the surgery route, I had three trials offered.
1) a randomised decision for lymph node removal or not with a three year follow up.
- not going to join that one, I’d prefer an informed decision about something as major as lymph node removal and not just let a computer randomly decide
2) an additional “metabolic” MRI scan for the MISSION study to see if they can get MRIs more accurate and reduce biopsies
- am definitely going for that one. Just an additional MRI which will be checked against the removed tissue. So nothing invasive.
3) a trial testing an experimental drug from Astra zenica, which is actually aimed at late stage cancer but they are assessing effects of short term treatment in localised tumours that will be removed
- still on the fence with this one. Seems relatively minor risk and the short term side effects are minimal. But need a think.
Has anyone else been offered these? What are you going to do?

Before any decision we would make we would read further into each and then make an informed decision .
You haven’t said what stage your PC is at . My OH is not on a curative pathway so any trial dealing with lymph nodes we would be inquisitive as he only has one inquinal lymph node outside his pelvis that they won’t treat . The other lymph node is within the pelvis .
take time and read up any literature on the trials before you make any decision

Hi Fzh2p0 and welcome. Can you clarify what the Astrazeneca trial is and this might help you get a response from someone who is on that particular drug. I have done a quick trawl and it looks as if it might be the ASCERTAIN trial (NCT05938270) which is assessing either an already approved hormone therapy (Darolutamide), a new PARP inhibitor or a combination of the two. Any trial comes with a risk and possible side effects which only you can decide whether you want to participate but make sure you understand what you are being asked to take and for how long.
That’s the one. Darolutamide, Saruparib or both.
I used to work in the clinical trials industry and have asked around my contacts and the feedback has been generally positive. Especially as the short dosage time means the side effects are minimal.
It’s just new being on this side of the trials fence. So was wondering if anyone else has been thinking of taking part in anything.
Its a phase 1 trial, but Saruparib has been around for a few years now, so not a very early high risk part of the phase
Got loads of Literature and am quizzing contacts in the clinical trials industry. It’s just all a bit daunting.
I have localised PC which the surgery team think can be fully removed. But as it’s Gleason 5+4 they need to get on it fast and have potential trials that may benefit.
Definitely not leaving lymph node to chance , that a discussion with the consultant. The MRI is fine, and the AZ drugs I’m tending towards yes.

I presume you have done your research into the risk of recurrence with a Prostatectomy depending on initial staging and type of procedure. From my understanding the PARP inhibitors boost the immune system whereas the Darolutamide is a hormone therapy which rapidly reduces testosterone levels.
We have a few who have been on the PACE NODES trial SBRT or are currently being offered a variation of it.
Also PEARLS trial.
Pearls trials with extended lymph node radiotherapy.

I’ve restarted looking for a trial re the inquinal lymph nodes we would. This will allow me to challenge the Oncologist as we mean business to try for a curative pathway.

We are fine thanks Liz but radiotherapy to the thigh starts on Monday now and probably for 10 days. All is organised for our 'holiday' in Athens. I am still trying to drill down the what next to deal with the rest of the mets.
